Usually yes for length, but not when removing them changes the meaning. A slug reading how-to-cook-rice is better than how-cook-rice, and the difference in length is not worth the awkwardness.
Hyphens or underscores?
Hyphens. Google treats a hyphen as a word separator and an underscore as a joiner, so word_two reads as one token.
Does changing a slug hurt?
Changing the slug of a page that already ranks means a new URL, and you need a 301 redirect from the old one or you lose everything it had earned. Get it right before publishing rather than tidying it later.
